Tropism Regeneration of Peripheral Nerves: A Novel Direction of the Treatment of the Severe Nerve Injuries

To discover injured peripheral nerve reproduction direction and thus provide new progress in the restoration of these injured peripheral nerves. Five SPF-grade rats was included in this study, whose left rear feet were used for experiment group and right rear feet were used for control group. Chitin sheath was used for sewing peroneal nerve's near-end and tibial nerve's far-end together, while peroneal nerve's far-end and tibial nerve's near-end were left untreated. Measure the conduction velocity and compound action potential of peroneal nerve, tibial nerve and sural nerve and weigh wet weight of gastrocnemius and tibialis anterior muscle in 12 weeks. Count the number of nerve fibers in peroneal nerve tibial nerve and sural nerve which were lately dyed by osmic acid. We found that the functional exponent of peroneal nerve in experiment group was -103.38±12.59, tibial nerve -80.37±10.58. Significant differences were found in wet weight of sural nerves between the experiment group and control group which were separately 0.87±0.09g and 1.71±0.06g (P<0.05). The conduction velocity of peroneal nerve was 15.76±1.72m/s in experiment group and 73.39±10.24m/s in control group, between which significant differences were found (P<0.05). The conduction velocity of tibial nerve in experiment group and control group was separately 12.23±0.80m/s and 69.21±9.93m/s and significant differences was also found (P<0.05). The amplitude of peroneal nerve's complex action potential (NCAP) was separately 25.66±1.44mV and 18.89±3.20mV in experiment and control group, and these of tibial NCAP was respectively 7.29±0.95mV and 17.20±2.25mV in experiment and control group, both in which significant differences were found(P<0.05). The number of tibial nerve sheathed fibers was respectively 20.81.80±98.81 and 4041.2±138.39 in experiment and control group, and the number of common peroneal nerve sheathed fibers was 1630±59.25 and 1068±59.08 respectively in those two groups ,and these of sural nerve sheathed fibers was 615.60±24.50 and 854.6±24.83 separately in those two groups, all of which presented significant differences (P<0.05). This study found that the tropism-distal-stumps regeneration was a pivotal character of peripheral nerves and the tropism power is big enough to make the newborn axons breaking through the physical sheath in rats.
